Distillation extraction VS CO2 supercritical extraction of lavender essential oil antioxidant effect

Foreword

The laboratory of the University of China used steam distillation and CO2 supercritical extraction to extract lavender essential oil to compare its anti-oxidant effects in vitro.

Method: Through DPPH, hydroxyl radical, superoxide anion, lipid peroxidation and hemolysis reaction system, detect and compare the antioxidant capacity of lavender essential oil extracted by the two methods.

Results: The lavender essential oil extracted by the two methods had strong antioxidant capacity and was dose-dependent. In the five oxidation systems, the antioxidant effect of lavender essential oil extracted by supercritical CO2 extraction method is stronger than that of lavender essential oil extracted by steam distillation method.

Conclusion: Lavender essential oil extracted by supercritical CO2 extraction method has stronger antioxidant capacity.

Testing process

DPPH•Determination of Scavenging Ability

The hydroxyl radical produced by the Fenton reaction reduces the absorbance of o-phenanthroline, the scavenger removes •OH, and the absorbance increases. The change in absorbance A before and after the reaction is used to determine the removal rate.

The lavender essential oil extracted by the two methods has a certain ability to remove •OH, and is in a dose-dependent relationship. The t test shows that the supercritical CO2, the lavender essential oil extracted by the extraction method has a strong ability to remove the .OH, and the steam distillation method extracts The antioxidant capacity of essential oils varies significantly.

Superoxide anion scavenging capacity measurement

The experimental results show that the lavender essential oil extracted by the two methods has an inhibitory effect on the generation of superoxide anions, and the effect increases with the increase of the dose.

The effect of lavender essential oil extracted by supercritical CO2 extraction method is stronger, EC50 is 60.5mg/L; the effect of lavender essential oil extracted by steam distillation method is weaker, EC50 is 72.5m/L, the difference is statistically significant (P<0.01).

Determination of Inhibition Ability of Egg Yolk Lipid Peroxidation

The lavender essential oil extracted by the two methods showed a good dose-effect relationship in the inhibitory effect of the yolk lipid peroxidation within the experimental concentration range.

Lavender essential oil extracted by supercritical CO2 extraction method has a significantly stronger inhibitory effect on yolk lipid peroxidation than that extracted by steam distillation method (P<0.01).

Red blood cell hemolysis test

The lavender essential oil extracted by the two methods has an inhibitory effect on red blood cell hemolysis. Within the experimental concentration range, the greater the concentration of the essential oil, the stronger the inhibitory effect. The lavender essential oil extracted by supercritical CO2 extraction has stronger anti-erythrocyte hemolysis effect than steam distillation. Extracted lavender essential oil (P<0.05).
