About coleus

Coleus forkskohlii belongs to the Lamiaceae – lavender family.

Coleus forskohlii is a member of the mint and lavender family (lamiaceae) that has seen centuries of use in Ayurvedic medicine.

This species is a perennial herb with fleshy, fibrous roots that grows wild in the warm sub-tropical temperate areas of India, Myanmar and Thailand. The essential oil is obtained from the fresh root material by hydrodistillation. One of its many components, sesquiphellandrene, provides a pleasant, woody aroma and is valued for its diverse biological benefits.

This “power” herb has an active ingredient know as forskolin.
Coleus has been utilized in ayurvedic medicines for many years.
Forskolin’s most common mechanism of action is that it increases the amount of cyclic AMP (adenosine monophosphate) in cells by activation of an enzyme called adenylate cyclase.
Cyclic AMP (cAMP) is one of the most important secondary messengers in the cell. It is regarded as the most crucial cell regulating compounds.

Forskolin

Under normal circumstances, cAMP builds by adenylate cyclase activation due to hormonal stimulation at the cell receptor site. However, forskolin seems to bypass this reaction and allows for an increase in intracellular cAMP to occur.

Why is it important to increase cAMP levels?

Well, this benefits athletes. It induces relaxation of the arteries lowering blood pressure, raised insulin secretion (which can help drive carbohydrates and protein into muscle cells for energy and recovery), increased thyroid hormone function (which can help raise metabolic rate), and significantly increase lipolysis (fat burning).

Forskolin also benefits different cellular enzymes. The breaking up of fat for fuel (lipolysis) is actually regulated by cAMP. Forskolin enhances lipolysis and also inhibits fat storage.

This is very beneficial for individuals attempting to reduce bodyfat and get lean. Forskolin also allows fat loss to occur by stimulating thyroid hormone production, which controls metabolic process and can increase metabolic rate, which can result in additional fat loss.

Forskolin also stimulates digestive enzymes, which aid digestion. They are shown to increase nutrient absorption in the small intestine.

Forskolin has been shown to be safe and effective and can be used as a sports supplement.

Supercritical CO2 extraction of Coleus Oil

Supercritical CO2 extraction is a popular method for extracting Coleus oil from the Coleus forskohlii plant. Coleus oil is known for its high content of forskolin, a bioactive compound that has attracted significant attention in the pharmaceutical and supplement industries. Supercritical CO2 extraction offers several advantages for extracting Coleus oil, including its ability to selectively extract target compounds while minimizing thermal degradation and solvent residue.

CO2 Extraction Process of Coleus Oil

The process of supercritical CO2 extraction of Coleus oil typically involves the following steps:

  • Preparation of Coleus Plant Material: The Coleus forskohlii plant is cultivated and harvested. The harvested plant material, usually the leaves or roots, is then dried and prepared for extraction. Proper drying helps maintain the quality and potency of the plant material.
  • Loading the Extraction Vessel: The prepared Coleus plant material is loaded into an extraction vessel or chamber that can withstand high pressures. The vessel is sealed, creating a closed system.
  • Extraction: Supercritical CO2 is introduced into the extraction vessel, where it acts as a solvent to extract the target compounds, such as forskolin, from the Coleus plant material. Under the specific temperature and pressure conditions, the supercritical CO2 penetrates the plant matrix and selectively dissolves the desired compounds, leaving behind unwanted components.
  • Separation: After the extraction process, the supercritical CO2 containing the dissolved compounds is transferred to a separation unit. Here, the pressure is reduced, causing the CO2 to revert to a gas phase. The Coleus oil, which is insoluble in gas-phase CO2, separates from the CO2 and is collected.
  • Post-Processing: The collected Coleus oil may undergo further refining processes, such as filtration or distillation, to remove any residual impurities and obtain a pure and high-quality oil with a standardized forskolin content.

Benefits of Supercritical CO2 Extraction for Coleus Oil

  • Selective Extraction: Supercritical CO2 extraction allows for selective extraction of the desired compounds, such as forskolin, from the Coleus plant. By adjusting the temperature and pressure, the extraction parameters can be optimized to target specific compounds while minimizing the extraction of unwanted components.
  • Preservation of Bioactive Compounds: Supercritical CO2 extraction maintains the integrity and potency of bioactive compounds in Coleus oil. The low temperatures used in the process help prevent thermal degradation, ensuring that the extracted oil retains its medicinal properties and forskolin content.
  • Solvent-Free and Safe: Supercritical CO2 extraction is a solvent-free process that utilizes carbon dioxide as the extraction medium. CO2 is non-toxic, non-flammable, and generally recognized as safe for use in food and pharmaceutical applications. This eliminates concerns about residual solvent contamination in the final Coleus oil.
  • Environmentally Friendly: Supercritical CO2 extraction is considered environmentally friendly due to its reduced use of organic solvents and minimal waste generation. CO2 used in the extraction process can be easily recycled and reused, contributing to a more sustainable extraction method.
